Cloning and expression analysis of <italic>CYP</italic>302<italic>a</italic>1 from <italic>Scylla paramamosain</italic>

In arthropods, periodic molting plays an important role in the growth, reproduction and morphogenesis. Periodic ecdysis is affected by a variety of environmental own factors, most which controlled concentration steroid hormones, ecdysone (Ecd) whose mainly active form 20-hydroxyecdysone(20E) hormone. Moreover, ecdysteroids play roles regulating development reproduction, CYP302al key enzyme biosynthesis ecdysteriods. We obtained full-length CYP302a1 cDNA sequence from mud crab Scylla paramamosain firstly, was named as Sp-CYP302a1 present study. The length 3 032 bp, with predicted 1 617 bp open reading frame (ORF) encoding 538 amino acids molecular weight 61.14 kDa, 5′UTR 205 3′UTR 210 bp. And putative protein contained 5 conserved domains including helix-C, helix-K, helix-I, PERF heme-binding. we found that there were no signal peptide sequences transmembrane Signal 5.0 Server TMHMM. phylogenetic tree showed S. paramamosain clustered Portunus trituberculatus Penaeus vannamei, then together other species. acid deduced CY302A1 vannamei into small branch, species, CYP306A1, CYP307A1, CYP315A1, CYP314A1 CYP18A1 encoded members P450 genes. analyzed expression pattern real-time PCR (qRT-PCR). results transcript detected Y-organ (YO) far exceeded tissues both males females. during larval development, kept fluctuating zoea I to second juvenile stage highest Z3 stage. Furthermore, process, levels Yo extremely low postmolt (stages A B), began increase reached peak at pre-molt (stage D), declined. this study, CYP302al gene cloned identified through analysis, combined tissue distribution, spatiotemporal expression, cycle changes. hypothesized might vital paramamosain.
